Comparison between different pretreatment methods to obtain fermentable sugars from amaranth stubble

Agro-industrial residues are gaining increasing importance for their use in various fermentation processes, including the production of substances with the biological activity of industrial value. Fermentable sugars (FS) are a key group of compounds for these processes. However, a series of steps must be carried out to facilitate their release. Pretreatment is a crucial step in obtaining these compounds. Therefore, the objective of this research work was to find a suitable method for delignification of amaranth stubble and subsequent production of fermentable sugars. For this purpose, five pretreatment methods were tested, which were classified as follows: physical (I. steam and II. ultrasound), chemical (III. 3.5N NaOH), and combined (IV. 3.5N NaOH + steam and V. NaOH (2, 2.5, 3, 3.5 N) + ultrasound). The Miller method (DNS) for reducing sugars was used to determine the concentration of FS, which would indicate the degradation of lignocellulose. This was corroborated by FTIR analysis. The methods that obtained the highest concentration of FS were the physical pretreatment (II. ultrasound) and the combined pretreatment (V. 3.5 N + ultrasound). Once the pretreatment method was defined, the saccharification process was carried out through conventional enzymatic hydrolysis and ultrasound-assisted hydrolysis, with the latter showing greater efficiency.
